In Borderlands: The Pre-Sequel!, Jack (also known as John) begins as a low-ranking programmer who obtains command of the Helios station despite animosity from CEO Harold Tassiter. After the Lost Legion invades Helios, Jack and six hired Vault Hunters evacuate to Pandora's moon, Elpis. To retake the station from Colonel Tungsteena Zarpedon, Jack initially spares the corrupt Meriff of Concordia but kills him after the Meriff shoots at his back, leading Jack to decide that mercy is a weakness. He creates a robot army using AI Felicity as its core until she must be destroyed for becoming unwilling to serve as a weapon. With aid from Mad Moxxi, Roland, and Lilith, the group retakes Helios and kills Zarpedon. However, at the Elpis Vault, Roland and Lilith betray Jack on Moxxi's advice; they destroy the Eye of Helios and nearly kill him. Inside the Vault, Jack uses an ancient artifact to learn of a powerful monster sealed in Pandora that will obey whoever unseals it.

In Borderlands 2, Handsome Jack introduces himself by blowing up the train carrying the player characters after luring them to Pandora with the goal of finding the next Vault. Throughout the game, he taunts the players via radio, similar to GLaDOS, while his relationship shifts from viewing them as irrelevant to hating them. After the mysterious Guardian Angel reveals she is working for Jack and sabotages Sanctuary's defenses, the player learns Angel is Jack's daughter. She helps the Vault Hunters destroy Eridium injectors keeping her alive, leading to her death at their hands. In revenge, Jack mutates Mordecai's bird Bloodwing, kills her after a boss fight, kills Roland, and kidnaps Lilith. Finally, in the Vault, Jack controls a monster to fight the players before being killed by either the Vault Hunters or Lilith.
